With the 96th overall choice in the third round of the 2019 NFL Draft, the Buffalo Bills picked Knox. On June 14, 2019, Knox penned his first-year contract with the group. In Week 3 against the Cincinnati Bengals on September 22, 2019, Knox scored his first NFL touchdown on a pass from Josh Allen.

He contributed to the Bills’ game-winning drive by grabbing a second Allen reception for 49 yards and outrunning two Bengals defenders en route to setting up Frank Gore’s touchdown run. The Bills prevailed 21-17 thanks to Knox’s three receptions for 67 yards and a touchdown. Knox’s first season was inconsistent, although he did manage to grab 28 of his 50 targets for 388 yards and two touchdowns.
